Melbourne's tram route 1 starts from the corner of Bell and Nicholson Sts East Coburg, where the terminus is in a reservation with a single track and waiting shelter. Upon departure, trams travel along Nicholson St past the former Coburg depot, now used by the Tramway Overhead Dept., then arrive at the Moreland Rd corner, where route 22 from Moreland joins, having travelled from Brunswick Depot in Cameron St next to the Upfield railway line. Route 22 crosses route 19 from North Coburg at Sydney Rd on its way to the junction, which is triangular to enable access to Brunswick Depot from Nicholson St.

Both routes travel along Holmes St through a largely residential area until a reverse curve at Albion St takes them into Lygon St and a light industrial area, passing Brunswick Rd which has a little used siding which used to be part of the Holden St line. The trackbed of the former Inner Circle railway is crossed at Park St, then trams go past the Melbourne General Cemetery and a Housing Commission estate before arriving at Elgin St Carlton where trams turn right. Fanciers of Italian cuisine can leave the tram here and walk further along Lygon St to find an area full of restaurants, shops and cafes all with a distinctive Italian flavour.

A little further along Elgin St, trams turn left into Swanston St and encounter the Melbourne University terminus, which caters for St Kilda Rd tram services. The university tram terminus is laid out with three tracks, routes 1 and 22 using the outer tracks and St Kilda Rd services using the middle track and two crossovers to shunt onto the citybound track.

Trams travel along Swanston St past the Royal Women's Hospital in Grattan St and enter the CBD at Victoria St opposite the City Baths, where the former Victoria St terminus and signal box can be seen on the right, as well as a double track connection to Elizabeth St. Travelling down Swanston St, LaTrobe St (routes 23,24,30,34 and the City Circle tourist tram) Bourke St ( route 86 to Bundoora RMIT and route 96 between St Kilda Beach and East Brunswick), Collins St ( route 11 to West Preston, route 12 to Sth Melb and St Kilda Beach and route 109 between Mont Albert and Port Melbourne) and Flinders St ( route 48 to North Balwyn, route 70 to Wattle Park, route 75 to East Burwood and the City Circle) are all crossed.

Federation Square can be seen on the left and Flinders St station on the right as trams head up St Kilda Rd crossing Princes Bridge over the Yarra River and passing the Alexandra Gardens on the left and the Victorian Arts Centre and the National Gallery on the right before arriving at Southbank Boulevarde, where route 22 terminates.

Turning right, trams pass a crossover and turn left into Sturt St with its plethora of car dealerships. CityLink's Sturt St interchange is a little further on as route 1 trams arrive at Kingsway, the Saturday route 4 terminus. Connecting curves are provided into Kingsway for the use of St Kilda Rd services 5,6,8,16 and 72 when they need to be diverted from Swanston St. Crossing Kingsway and route 55 between Domain Rd interchange and West Coburg, trams enter Eastern Rd and turn right into Park St Sth Melbourne before arriving at Clarendon St and the route 12 crossing.

Leaving Clarendon St and its strip shopping centre, Park St is followed, crossing the St Kilda Light Rail Line ( route 96) between Sth Melbourne and Albert Park stations then trams turn left into Montague St Albert Park past the St Vincent Gardens before turning right into Bridport St and the Albert Park shopping centre, which is followed after turning left into Victoria St to the terminus at Beaconsfield Parade, a short walk to South Melbourne Beach. Tram travellers can take their skateboards and roller blades along with them as there is an excellent bike and skate trail along the foreshore near the tram terminus, which is in reservation containing the necessary single tram line, seats and shelter.
